چکیده

The Solute Carrier 15 (SLC15) family of peptide transporters, alias H+-coupled oligopeptide cotransporter family, is a group membrane transporters known for their key role in the cellular uptake di- and tripeptides (di/tripeptides). Of its members, SLC15A1 (PEPT1) chiefly mediates intestinal absorption luminal di/tripeptides from overall dietary protein digestion, SLC15A2 (PEPT2) mainly allows renal tubular reuptake ultrafiltration brain-to-blood efflux choroid plexus, SLC15A3 (PHT2) SLC15A4 (PHT1) interact with both histidine, e.g. certain immune cells, SLC15A5 has unknown physiological function. In addition, SLC15 variably interacts very large number peptidomimetics peptide-like drugs. It conceivable, based on currently acknowledged structural functional differences, to divide into two subfamilies [3].

Functional expression of SLC15 peptide transporters in rat thyroid follicular cells.

Peptide transport and expression of SoLute Carrier 15 (SLC15) peptide transporters was assessed in rat thyroid tissue and a rat thyroid cell line (PC Cl3 cells). Peptide transport was studied by monitoring the uptake of the fluorophore-conjugated dipeptide beta-Ala-Lys-N(epsilon)-7-amino-4-methyl-coumarin-3-acetic acid (Ala-Lys-AMCA). Expression of SLC15-specific mRNA transcripts was analyzed b...

A POTluck of peptide transporters.

The uptake of diet-derived peptides is mediated by the conserved proton-dependent oligopeptide transporter (POT) family. Crystal structures of bacterial transporters in the inward-open conformation and in an occluded conformation published in The EMBO Journal provide structural insight into the proton-driven peptide transport cycle. Lipid bilayers are generally not permeable to amino acids, pep...

Proton movement and coupling in the POT family of peptide transporters

POT transporters represent an evolutionarily well-conserved family of proton-coupled transport systems in biology. An unusual feature of the family is their ability to couple the transport of chemically diverse ligands to an inwardly directed proton electrochemical gradient. For example, in mammals, fungi, and bacteria they are predominantly peptide transporters, whereas in plants the family ha...

Symmetry and Structure in the POT Family of Proton Coupled Peptide Transporters

The POT family of proton coupled oligopeptide transporters belong to the Major Facilitator Superfamily of secondary active transporters and are found widely distributed in bacterial, plant, fungal and animal genomes. POT transporters use the inwardly directed proton electrochemical gradient to drive the concentrative uptake of diand tri-peptides across the cell membrane for metabolic assimilati...

The SLC2 (GLUT) family of membrane transporters.

GLUT proteins are encoded by the SLC2 genes and are members of the major facilitator superfamily of membrane transporters. Fourteen GLUT proteins are expressed in the human and they are categorized into three classes based on sequence similarity. All GLUTs appear to transport hexoses or polyols when expressed ectopically, but the primary physiological substrates for several of the GLUTs remain ...
